Which measurement should I take first?

Work out the roof pitch first — it determines the true (sloped) surface area from a flat footprint measurement, and every shingle estimate depends on getting that multiplier right. Once you have pitch and footprint, the shingle calculator converts squares into bundle counts, factoring in waste for hips, valleys, and starter courses.
